Abstract

Effects adenosine receptor agonists, antagonists and calcium channel blocker on naloxone-induced jumping and diarrhea in morphine-dependent mice were studied. The adenosine A1 receptor agonist (CCPA: 0.5 mg/kg i.p.), and A2a receptor antagonist (SCH58261:1mg/kg i.p.) significantly reduced jumping. CCPA also reduced diarrhea significantly but SCH58261 did not affect it. calcium channel blocker (felodipine: 2,5,5,10 mg/kg i.p.) decreased jumping and diarrhea .but the effect of 5 mg/kg doses of felodipine, on reducing of symptoms were significant. But dose-dependency was not seen. In combination therapy mice treated with felodipine 5 mg/kg and CCPA or SCH58261 before naloxone. Results of combination therapy with felodipine (5mg/kg) and CCPA or SCH58261 on jumping and diarrhea were also significant. Our results demonstrated that A1 and A2a adenosine receptors mediate the effect induced by adenosine in opiate withdrawal syndrome. Ca2+ channel blockers suppress the behavioural expressions of morphine withdrawal. Using Ca2+ channel blockers in combination with adenosine receptor agonists and antagonists decreased morphine withdrawal symptoms. But the potentiating effect was seen just in felodipine and SCH combination therapy.
