Abstract
The 5-Br-meso-VO-Salen, The VO-Salen and The Salen complexes were synthesized in The laboratory. the evaluation of free radical scavenging of Salen using DPPH assay showed that Salen was a least effective antioxidant than its vanadium oxide complexes. The biological and antiproliferative activity of The 5-Br-meso-VO-Salen, The VO-Salen and The Salen complexes were evaluated by MTT reduction assay on LNCaP Prostate cancers cell line. Treating LNCaP cells with various concentrations of the three complex, showed that the growth of the cells were inhibited with IC50 of 403.55, 428.42 and 736.276 µM by The 5-Br-meso-VO-Salen, The VO-Salen and The Salen complexes, respectively. Morphological studies of thereated cells indicate the occurrence of necrotic cell death. In order to evaluation of programmed cell death, quantitative expression of the genes which encode apoptotic cell signaling factores like caspase 3, bax and bak were evaluated after treating by the 5-Br-meso-VO-Salen and the VO-Salen complexes. The data analysis showed that the caspase 3 gene expression was increased by 7.16 and 1.82 fold , using the 5-Br-meso-VO-Salen and the VO-Salen complexes, respectively. The VO-Salen and the 5-BrMVO-Salen induce bak and bax genes expression respectively which lead to caspase3 gene expression and ultimately triggers mitochondrial apoptotic pathway. caspase3 gene expression can be occurred by external pathway of apoptosis with ligation of this complexes in receptors.

Persian Abstract: | در اين پژوهش کمپلکس معدنيVO-Salenو 5-Br-meso-VO-Salenاز تراکم آلدولي اتيلن دي آمين سالسيل آلدهيد در حضور ترکيب واناديوم استيل استونات سنتز گرديد. ابتدا قدرت آنتي¬اکسيداني Salen به وسيله¬ي آزمون DPPHمورد ارزيابي قرار گرفت، نتايج نشان داد که Salen قدرت آنتي¬اکسيداني بسيار ضعيفي در مقايسه با کمپلکس¬هاي واناديوم اکسيد آن دارد. ارزيابي اثرات سميت سلولي و ضد تکثيري کمپلکس¬هاي Salen، VO-Salenو 5-Br-meso-VO-Salenنشان داد که رشد سلول¬هايLNCaP بعد از 16 ساعت تيماربه ترتيب با IC50 برابر با 276/736 ، 42/428 و 55/403 ميکرومولار مهار گرديد. جهت ارزيابي رويداد مرگ برنامه¬ريزي شده¬ي سلولي، بيان کمي برخي از ژن¬هاي کد کننده¬ي فاکتورهاي مسير سيگنالينگ مرتبط با آپوپتوزيس سلولي مورد بررسي قرار گرفت يافته¬هاي حاصل از اين آزمايش¬ها نشان داد هر دو کمپلکسVO-Salenو 5-Br-meso-VO-Salen بيان کاسپاز 3 را به ترتيب به ميزان 82/1 و 16/7 برابر القا مي¬کند، القاي بيان ژن bak توسط VO-Salenو نيز بيان ژن bax به¬وسيله¬ي 5-Br-meso-VO-Salen ممکن است باعث پليمريزاسيون پروتيئن¬هاي Baxو Bakدر غشاي ميتوکندري شده و با بيان کاسپاز3 مسير ميتوکندريايي آپوپتوزيس ادامه يابد. | ||||
