Abstract
Acute leukemia of promyelocyte is one of the most prominent types of acute leukemia with the displacement of chromosomes 15 and 17, which do not have the ability to convert to adult cells. In recent years, the treatment of this type of immature cancer has been used in a new method of differentiation by differentiating agents. The aim of this study was to investigate the effect of Nigella sativa Alcoholic Extract and Vitamin E on the differentiation of HL-60 cancer cells. In this experimental study, the HL-60 cancer cell line was prepared from the Institute of Pasteur Institute of Iran. In order to determine the cytotoxic effects of vitamin E and Nigella sativa Alcoholic extract on HL-60 cells separately and combined, these cells were treated with different concentrations of vitamin E and Nigella sativa Alcoholic extract for 24 hours. Subsequently, cell survival Determined by MTT. In addition, differentiation was made on this cell line. The results showed that Nigella sativa Alcoholic extract at 650 μg / ml concentration in combination with 15 μg / ml vitamin E increased its anti-proliferative and differentiative potency during 24 hours.

| Persian Abstract: | لوسمی حاد پرومیلوسیتی یکی از انواع لوسمی های حاد با جابجایی کروموزوم 15 و 17 است که توانایی تبدیل به سلول های بالغ را نداشته ودائماً تکثیر می شوند. در سال های اخیر جهت درمان این نوع از سلول های سرطانی نابالغ از روش جدید تمایز درمانی به وسیله عوامل تمایز دهنده استفاده می شود . هدف مطالعه حاضر بررسی اثر عصاره سیاه دانه و ویتامین E بر روی تمایز سلول های سرطانی HL-60 بود. در این مطالعه ی تجربی در محیط آزمایشگاهی، رده سلولی سرطان HL-60 از موسسه انیستیتو پاستور ایران تهیه شد . به منظور تعیین اثرات سیتوتوکسیک ویتامین E و عصاره الکلی سیاه دانه بر روی سلول های HL-60 به صورت جداگانه و توام، این سلول ها با غلظت های مختلف ویتامین E و عصاره الکلی سیاه دانه به مدت 24 ساعت تیمار شدند .سپس میزان بقاء سلولی توسط سنجش MTT تعیین شد. بعلاوه ایجاد تمایز روی این رده ی سلولی نیز بررسی شد . نتایج به دست آمده نشان داد عصاره الکلی سیاه دانه در غلظت µg/ml 650 در ترکیب با غلظت µg/ml 15 از ویتامین E در طول 24ساعت قدرت تمایزی و ضد تکثیری آن را افزایش داد . |
