Abstract

The fungus Diplodia bulgarica, that belongs to the family Botryosphaeriaceae and has been described in recent years, is the causal agent of canker in various trees. This pathogen is considered as an important agent causing canker on apple trees in West Azarbaijan province but, little information is available on its distribution in central regions of the province. In this research, sampling was performed from apple orchards located in Urmia, Oshnavieh, Mahabad, Salmas, Mihammadyar and Miandoab, and fungal isolates were isolated from infected plant tissues and were then purified. Identification of the isolates were first carried out based on morphological characteristics and then using phylogenetic analysis of ITS (Internal transcribed spacers) and TEF1-α (Translation elongation factor 1-α) sequences. In total, 105 fungal isolates were obtained that 100 isolates were identified as D. bulgarica and the rest of them belonged to Cytospora parasitica, C. cincta and Clonostachys rosea The species D. bulgarica was the predominant pathogen and was distributed intensively across the studied regions however, incidence of the disease was high in the vicinity of Urmia compared with other regions.

Persian Abstract: قارچ Diplodia bulgarica، که متعلق به خانواده Botryosphaeriaceae می¬باشد و در سال¬های اخیر معرفی و توصیف شده است، عامل ایجاد شانکر در درختان مختلف است. این بیمارگر به عنوان عامل مهم ایجاد شانکر در درختان سیب در استان آذربایجان غربی در نظر گرفته می¬شود ولی اطلاعات محدودی در زمینه پراکنش آن در مناطق مرکزی استان وجود دارد. در این تحقیق، نمونه¬برداری از باغ-های سیب در ارومیه، اشنویه، مهاباد، سلماس، محمدیار و میاندوآب انجام گرفت و جدایه¬های قارچی از بافت¬های آلوده جداسازی و خالص¬سازی گردید. شناسایی جدایه¬ها ابتدا با استفاده از مشخصات ریخت¬شناختی سپس با تجزیه نسب¬شناختی توالی¬های ITS (Internal transcribed spacers) و TEF1-α (Translation elongation factor 1-α) انجام گرفت. در مجموع 105 جدایه قارچی به دست آمد که 100 جدایه به عنوان D. bulgarica شناسایی شدند و بقیه متعلق به Cytospora parasitica، C. cincta و Clonostachys rosea بودند. گونه D. bulgarica بیمارگر غالب بود و به طور وسیعی در همه مناطق مورد بررسی پراکنده بود ولی شیوع بیماری در اطراف ارومیه بیشتر از سایر مناطق بود.
